Trying to assign flaps to joystick buttons but flaps lever does not move (744) only vibrates but remains stationary.

That is strange, as if some axis is continuously resetting your inputs.
Make sure you don't have an axis assigned to flaps and also keep the copilot disabled or it will also try and move the flaps.
Can you move the flaps normal with the mouse wheel?
Mouse wheel? Ahhh. Didn’t realise you could. I also didn’t realise we had an copilot either. How does one enable / disable this function?
Can mouse wheels be user assigned? Mice are now pound/dollar store items and a few on a hub could be useful for trim axes via their cheap Gray code wheels.

Mouse wheel affects whatever you are pointing at in the cockpit, that's why there is no control assignment possible on the mouse wheel.
How do you know if the copilot has been enabled, or not?
The copilot interface is usually hidden but you can enable it in the settings with the "overlay". When the copilot is flying you have no manual control anymore (that's how you know) and I believe the default key is "c" for copilot.
When you just press "c" it holds your attitude and airspeed. If you press it again you regain control.
You can also use the autopilot disengage (or take over function) to become pilot in command.

There is no such thing. I have checked everywhere in settings menu.
Pressing C has no effect whatsoever. Apparently if you enable copilot then the flight is overtaken, including the takeoff.
I have only just downloaded from steam so assuming I have the updated version.
You need to assign some inputs in settings, I think I put A in myself for engage autopilot.
So whats C?
